The ImageStream X Mark II combines the features of standard flow cytometry and microscopy, enabling rapid and quantitative analysis of cells with the sensitivity of a cytometer and the spatial resolution of a microscope (at 60× magnification, the pixel size is 0.3 × 0.3 µm).
The system available in the facility is equipped with two excitation lasers (488 nm and 642 nm), a TDI CCD camera capable of capturing up to 6 images of each individual cell in flow, and an acquisition rate of up to 5,000 cells per second (including brightfield, side scatter (SSC), and four fluorescence channels).
Three objectives are available for image acquisition: 20× (0.5 NA), 40× (0.75 NA) and 60× (0.9 NA). Available bandpass filters include: 528/65 (Ch2), 577/35 (Ch3), 610/30 (Ch4), 702/85 (Ch5) and 762/35 (Ch6). The “Inspire” software (v.200.1) controls image acquisition in both brightfield and fluorescence modes, while “IDEAS” software (v.6.2) enables quantitative analysis of cell images and population-level statistical processing.
This instrument allows the analysis of several biological processes, including: translocation, co-localization, cell cycle, apoptosis, autophagy, vesicle internalization and other cellular characteristics.
